Three-day broccoli sprouts as a timed electrophile pulse

A metabolic hypothesis. N-acetylcysteine is not on the same pathway.

7 October 2026

Disclaimer. This is a working hypothesis, not medical advice and not a dosing recommendation. Glucoraphanin content of sprouts varies several-fold with seed, age, and storage. Conversion to sulforaphane depends on chewing and intact myrosinase. Human trials have used a wide range, from about 20 g of sprouts a day to very large sprout homogenates, with phase II induction discussed around 50–100 µmol sulforaphane equivalents. People with active cancer, especially KEAP1- or NFE2L2-altered tumors, should not treat Nrf2 activation as benign. Sulforaphane can inhibit CYP enzymes and may alter drug handling. Iodine deficiency with goiter risk, anticoagulation, and pregnancy are separate reasons to clear a crucifer load with a clinician. N-acetylcysteine has its own indications and adverse effects and is not interchangeable with food. Food-level sprout use in healthy adults has looked well tolerated in short trials. That is not a safety claim for chronic high intake.

Hypothesis

A small raw serving of 3-day broccoli sprouts, taken at the first meal on repair days only, raises a transient Keap1–Nrf2 defense program that supports the mitochondrial engine without occupying the weekly growth window. The molecule is cleared within a day. The enzyme program lasts one to two days and is allowed to fall before the leucine meal. N-acetylcysteine is not a substitute for that pulse and is not a standing input beside it.

The sprout is a signal, not a fuel. Fuel, circadian light, and resistance still do the work the crucifer cannot. The capsule of sulforaphane glucosinolate is a weaker travel spare of the same idea, and only if an active myrosinase source is eaten with it.

Form and conversion

Three-day sprouts are the right form because they carry both halves of the reaction. They concentrate glucoraphanin far above mature broccoli and still hold active myrosinase, provided they are not cooked. Chewing brings enzyme and glucosinolate together. Sulforaphane, the electrophilic isothiocyanate, is the signal. Glucoraphanin is stable and inactive.

Plant myrosinase converts reliably. Gut bacteria convert variably, often in a low range and sometimes near zero after antibiotics. A glucoraphanin capsule without mustard seed or a raw crucifer is that lottery. Heat kills sprout myrosinase. If the crucifer has been cooked, brown mustard powder or raw daikon after the pot restores the enzyme the heat destroyed.

A practical food serving in the range discussed in clinical work is about 20–40 g of fresh sprouts, chewed until the bite is hot. A couple of ounces of sprouts can match a large serving of market broccoli. Content is not standardized. Urinary sulforaphane metabolites, not the label weight, are the internal dose.

The defense track

Sulforaphane modifies reactive cysteines on Keap1, especially Cys151. Nrf2 escapes proteasomal destruction, enters the nucleus, dimerizes with a small Maf, and binds the antioxidant response element. The induced set is capacity, not scavenging: NQO1, heme oxygenase-1, both subunits of glutamate-cysteine ligase, glutathione synthetase, the glutathione S-transferases, UDP-glucuronosyltransferases, thioredoxin reductase, and several NADPH-generating enzymes.

Nrf2 is the cell’s electrophile sensor. Under quiet conditions Keap1 holds it and hands it to a Cul3 ligase, so the protein barely exists. The system is built to stay off until a thiol-reactive signal arrives. A molecule that is itself an antioxidant does not open it. A molecule that looks, briefly, like a toxin does.

Three cross-talks place the pulse on the repair arm. Nuclear Nrf2 competes with and indirectly restrains NF-κB, a tilt rather than a shutdown of host defense. The same program raises glutathione and NADPH, which the respiratory chain needs, and feeds forward into TFAM and mitochondrial content. In muscle-cell models a sulforaphane pulse shows nuclear Nrf2 within hours and higher respiration a day later. It is not a dedicated mitophagy signal. p62 can bind Keap1 and release Nrf2, and AMPK and Nrf2 cross-activate, so the sprout shares weather with the repair side of the week. It is not an AMPK drug.

The Janus face is the constraint on more. Inducible Nrf2 protects intact tissue. Constitutive Nrf2, from loss of KEAP1 or gain of NFE2L2, can support tumor growth, metabolic rewiring, and therapy resistance. Food-level, pulsed activation stays on the protective side only if the molecule is allowed to clear.

Molecule versus program

Plasma peak is about 1 to 1.5 hours after a myrosinase-active dose. Elimination half-life is about 1.8 to 2.5 hours for the parent and for the principal mercapturic metabolite, sulforaphane-N-acetylcysteine. Roughly 60 percent of a dose is in the urine by 8 hours, and excretion is essentially finished by 24 hours. Tissue metabolites follow the same curve.

What lingers is transcription. A brief pulse raises NQO1 and HO-1 mRNA for about 12 to 24 hours, and the proteins can stay elevated past 48 hours in cell models. Repeated mornings accumulate the enzymes. They do not accumulate the molecule. A day-7 serving is not still circulating on day 1. At most it leaves a fading phase II tail, which is defense, not an AMPK lock and not residual drug in the growth window.

Clock and week

Nrf2 abundance is shaped by BMAL1 and CLOCK and rises in the active phase, late morning to early afternoon in diurnal humans. Sulforaphane peaks in plasma one to two hours after the meal, so the serving belongs after morning light, at the first meal, inside the feeding window. Fasting itself nudges Nrf2, so breaking the fast with the sprouts stacks the signal. An evening serving puts an activity-phase electrophile into the melatonin window.

Human trials that moved phase II enzymes, pollutant excretion, or bowel habit used repeated daily exposure, including about 20 g of sprouts a day for several weeks, not a weekly hit. The molecule clears daily, so the cadence is daily on repair days. The weekly exception is the growth day, not a multi-day washout.

Skip day 1 if the signals are to stay clean. Use the sprouts on days 2–7. That avoids stacking an AMPK-leaning electrophile on the leucine meal. One skipped day is enough. Do not stretch the gap and expect the enzyme program to substitute for the next pulse.

N-acetylcysteine is not the same pathway

NAC and sulforaphane meet at glutathione and enter from opposite directions. NAC supplies substrate. The sprout induces the machinery that uses substrate. With adequate protein and sprouts on days 2–7, a standing NAC dose is unnecessary and can work against the week.

N-acetylcysteine is a cysteine prodrug. It is deacetylated to cysteine, and cysteine is the rate-limiting substrate for glutamate-cysteine ligase. NAC is a nucleophile and a reductant. It can break disulfide bonds, which is why it is mucolytic. It does not modify Keap1 the way an electrophile does. At sufficient concentration it does the opposite of sulforaphane: it buffers the thiol-reactive signal that would have released Nrf2.

Sulforaphane never donates cysteine. It raises transcription of glutamate-cysteine ligase, glutathione synthetase, and the glutathione S-transferases. The sprout builds the factory. Dietary cysteine and methionine, which an animal-protein meal already provides, stock it. NAC is a second stocking method, not a second signal.

They collide on training and repair days. The exercise pulse works by the same cysteine logic as the sprout: reactive species modify Keap1, Nrf2 rises, the defense set is written. High-dose antioxidants taken around the session can consume that signal before it is read. Intravenous NAC has blunted signaling in some fatigue models. Oral NAC is milder and less consistent. A short loading phase can help repeated high-intensity work under heat or hypoxia. Across a training block the adaptive effect is unsettled, and routine chronic use is not supported. Days 2–7 are supposed to carry a clean electrophile and training signal. A daily reductant on top of that flattens both.

Keep NAC for a defined job: mucus, an acetaminophen exposure, a clinician-directed course. Keep it off the mornings when the sprout and the session are the signal.

What this hypothesis does not claim

  • It does not claim that sprouts treat, prevent, or reverse disease.
  • It does not claim a standardized sulforaphane yield from a handful of sprouts.
  • It does not claim that a glucoraphanin capsule without myrosinase reproduces the food.
  • It does not claim that NAC is harmful in its clinical uses, only that it is the wrong standing input beside this pulse.
  • It does not claim that skipping day 1 is required by pharmacokinetics. It is pathway hygiene. The molecule would be gone either way.

What would count against it

  • No rise in urinary sulforaphane metabolites after a chewed serving means conversion failed.
  • No change in a phase II marker across days 2–7 means the pulse is too small.
  • Worse recovery, sleep, or training quality on sprout mornings means the dose has left hormesis.
  • A growth week that stalls only when sprouts are added to day 1 would support the skip.
  • A clear benefit from adding daily NAC on top of protein and sprouts would challenge the redundancy claim. That result is not the current expectation.

Working rule

Days 2–7: 20–40 g raw 3-day broccoli sprouts, chewed at the first meal, after morning light. Day 1: off. Mustard powder only if the sprouts are unavailable and the crucifer has been cooked. The glucoraphanin capsule is a travel spare, and only with an enzyme source. No scheduled NAC on the sprout and training mornings.

The rule is narrow on purpose. The sprout flickers a defense program. The week lets that program fall before the growth meal. NAC stocks a tank the meal and the induced enzymes already fill.

A working hypothesis, open to correction and revised as new evidence arrives. Last revised October 17, 2026.

See the notice at the top of this page. Working hypothesis, not medical advice and not a dosing recommendation. The serving and timing above are the essay’s own working rule, as written. Active cancer (especially KEAP1- or NFE2L2-altered tumors), drug handling through CYP enzymes, iodine deficiency with goiter risk, anticoagulation, and pregnancy are reasons to clear a crucifer load with a clinician. NAC has its own clinical uses; this page does not say to stop NAC a clinician has prescribed.
